GLP-1 (glucagon-like peptide-1) receptor agonists mimic the gut hormone GLP-1 to regulate blood sugar, suppress appetite, and slow gastric emptying. Compounded semaglutide became widely used during commercial product shortages of Ozempic and Wegovy.

How It Works

GLP-1 receptor agonists bind to GLP-1 receptors in the pancreas, brain, and gut. They stimulate insulin release, suppress glucagon, slow digestion, and reduce appetite by acting on hypothalamic satiety centers.

Why Compounded?

Ozempic and Wegovy shortages drove widespread demand for compounded semaglutide. Compounding also allows customized titration schedules, lower starting doses for sensitive patients, and combination formulations with B12 to reduce side effects.

Compounded semaglutide was permitted during FDA shortage periods. As of early 2025, the FDA removed semaglutide from the shortage list, restricting compounding by 503B outsourcing facilities. 503A pharmacies may still compound for specific patient needs.
